dc.descriptionIn order to identify members of the bZIPs family of proteins in Trypanosoma cruzi, gel shifts experiments were done using oligonucleotides with AP-l sequences. Using a double stranded probe, three specific complexes were formed, named A, B and C. When using a single stranded probe, only complexes A and C were obtained. The relative amount of the complex B (double stranded), increased as a function of length probe and ionic strength. UVC cross-linking generated 100, 60 and 39 kDa bands for complex A, 46 and 43 kDa bands for complex B and just one band of 43 kDa for complex C. The partial purification of the proteins confirmed the common nature of complexes B and C. A new gene from T. cruzi belonging to the CCHC zinc finger proteins family was cloned by PCR. The new protein contains seven zinc finger domains and was named poly zinc finger protein, PZFP-1. Southern blots experiments showed that the gene is arrayed in tandem copies. Northern and Western blots assays showed a strong expression in epimastigote forms. The new protein forms strong dimers and could bind to single stranded CCHC binding sites. Inmunofluorescence assays showed that the protein is mainly citoplasmatic, but is also present in discrete regions of the nucleous. Fractioning of epimastigote extracts showed that both AP-1 and CCHC DNA-binding activities, copurify, and that PZFP-1 could be involved in the formation of the complex B.-
