dc.description | El objetivo de este trabajo de tesis es estudiar la dinámica de los entornos sonda-caja de polímero en películas nanométricas de polímeros amorfos. Para ello se utilizaron sondas fluorescentes, altamente sensibles a las propiedades fisicoquímicas de su entorno (Rojo Nilo) embebidas en polimetacrilatos de alquilo. La emisión de la sonda se estudió a escala macroscópica (ensamble de moléculas de colorante) y a nivel de moléculas individuales. En ambos casos se analizaron las propiedades espectrales y rotacionales de la sonda en la caja de polímero. La espectroscopía de emisión de fluorescencia de moléculas individuales de Rojo Nilo mostró fluctuaciones espectrales cuya frecuencia y amplitud son independientes del espesor y tipo de polímero, mientras que las reorientaciones de la sonda dependen del espesor de la película, que esta asociado a cambios en el valor de la temperatura de transición vítrea por efecto del confinamiento. El fotoblanqueo se encuentra siempre presente en experimentos de este tipo, y con esta motivación, se propuso una solución al estudio de la dinámica rotacional del ensamble en presencia de fotoblanqueo bajo excitación linealmente polarizada. Se obtuvo así el coeficiente de difusión rotacional de la sonda en los polímeros, que explica la tendencia observada para la movilidad de moléculas individuales. Las medidas se realizaron con dos configuraciones de microscopio de fluorescencia: confocal con detección espectrográfica y de campo amplio con detección polarizada. dc.description | In this Thesis, we study the dynamics of probe-polymer cage environments in thin polymer films. For this purpose, we employed fluorescent probes with a high sensitivity to the physicochemical properties of its environment, as Nile Red. The selected polymer matrixes were the polyalkylmethacrylates. The distance and interactions between the probe and its near environment (polymer cage) affect some emission features of the dye. Examples of these features are: the emission spectrum and probe reorientation. Probe emission was studied at macroscopic (ensemble of dye molecules) and single molecule level. Spectral and reorientational properties of the emission were studied. At the single molecule level, fluorescence spectroscopy of individual Nile Red molecules shows spectral fluctuations whose frequency and amplitude are independent of the thickness and the polymer type. The rotational dynamics of single molecule in the ground state, followed by polarized emission measurements, showed a dependence of amplitude and frequency of reorientation events on polymer film thickness. A new method to study probe rotational diffusion, which combines anisotropy and photobleaching with linearly polarized excitation is presented. It is based on 1-photon photobleaching and isotropic rotation of the dye. The diffusion coefficient of Nile Red in the polymer matrix of different thickness was obtained and it was correlated to single molecule reorientations results. Two fluorescence microscopy configurations were used: confocal microscopy with spectrographic detection, and wide-field microscopy with polarized emission detection.
