dc.description | Las células NK son células del sistema inmune, críticas en la defensa contra células tumorales y células infectadas, que se activan luego de sensar señales a través de receptores activadores y por acción de citoquinas secretadas por macrófagos y células dendríticas (CD), entre las cuales IL-12 es considerada una de las más relevantes. Recientemente se han descripto nuevos miembros de la familia de IL-12, entre ellos IL-23 e IL-27. IL-23 posee efectos pro-inflamatorios, pero existen datos contradictorios en cuanto a su rol en el contexto tumoral. En cambio, IL-27 induce una potente respuesta anti-tumoral, pero posee efectos tanto pro- como anti-inflamatorios. Debido a que el efecto de estas citoquinas sobre las células NK humanas aún no ha sido investigado, el objetivo de esta tesis doctoral fue estudiar el efecto de IL-23 e IL-27 sobre estas células. En primer lugar, demostramos que IL-23 e IL-27 secretadas por CD contribuyen a estimular la producción de IFN-γ por células NK. Mediante el empleo de inhibidores farmacológicos, demostramos que la estimulación de células NK con las citoquinas recombinantes indujo la producción de IFN-γ a través de la activación de las vías de JNK, PI3K, MEK1/2, NF-κB y mTOR, y que IL-27 activó además a STAT-1. Observamos también que tanto IL-23 como IL-27 generaron un efecto de priming para IL-18, exhibiendo estas citoquinas un efecto sinérgico con IL-18 para la secreción de IFN-γ. En el caso de IL-27, dicho efecto involucró un aumento en la expresión de T-bet y del receptor de IL-18. A su vez, ambas citoquinas indujeron un aumento en la expresión de los marcadores de activación celular CD25 y CD69, y potenciaron la citotoxicidad celular dependiente de anticuerpos. IL-27, pero no IL-23, estimuló la respuesta citotóxica de las células NK de manera dependiente de NKp46 y de las vías de NF-κB y mTOR, a través de la vía secretoria y de TRAIL, pero no la de FasL. Detectamos además que IL-18 potenció la actividad citotóxica inducida por IL-27 a través de la inducción de un aumento en la expresión de ICAM-1 en las células blanco mediado por IFN-γ. En conjunto estos resultados indican que IL-23 e IL-27 estimulan las funciones efectoras de las células NK, lo que puede ser relevante durante situaciones tanto fisiológicas como patológicas. | - |
dc.description | NK cells are immune cells, critical during immunity against intracellular infections and tumors, that became activated after recognition through activating receptors and macrophage and dendritic cell (DC)-derived cytokines, among which IL-12 is considered to be one of the most relevant. New members of the IL-12 family have been recently described, including IL-23 and IL-27. IL-23 display pro-inflammatory properties, however its role during the anti-tumor immune response is controversial. On the other hand, IL-27 induces a potent anti-tumor immune response, but shows pro- and anti-inflammatory effects. The effect of these cytokines on human NK cells has not been yet studied, thus the objective of this doctoral thesis was to study the effects of IL-23 and IL-27 on human NK cells. We first demonstrated that DC-derived IL-23 and IL-27 induced an enhanced production of NK cell-derived IFN-γ through the activation of JNK, PI3K, MEK1/2, NF-κB and mTOR pathways. In addition, IL-27 also activated the STAT-1 pathway. We also observed that IL-23 as well as IL-27-primed NK cells for IL-18-induced IFN-γ secretion, showing a synergistic effect, which in the case of IL-27 was associated with upregulation of T-bet and IL-18 receptor expression. Moreover, both cytokines induced upregulation of the activation markers CD25 and CD69 and enhanced the antibody-dependent cell-mediated cytotoxicity. IL-27, but not IL-23, stimulated NKp46-dependent NK cell-mediated cytotoxicity through NF-κB and mTOR pathways. In addition, the TRAIL and secretory pathways, but not Fas-FasL interaction, are involved in this effect. We have also observed that IL-18 enhanced the IL-27-induced cytotoxic activity through the upregulation of ICAM-1 on target cells, mediated by IFN-γ. Together our results show that IL-23 and IL-27 stimulate NK cell effector functions, which may be relevant during different physiological as well as pathological situations. | - |
